Yes. The only thing that will be different is that if someone's draft clock expires, it won't automatically activate clock of the next player.

A few years ago I was part of a Madden online franchise in which each team was drafted from the league's current player pool. Not only was it a lot of fun but we got the draft done in a few hours.

So it's certainly possible.
