When asked by Collider if he [Damon Lindelof] would be writing the script, he said: "I am not. Ridley [Scott] and I talked at great length during the story process of the first movie about what subsequent movies would be if Prometheus were to be successful. And he was like, 'Well, okay, it’s not like I asked you anyways.' He and I are on excellent terms and it was a dream come true to work with him. But much to the delight of all the fanboys, I don’t see myself being involved in Prometheus-er."

Yes, it is a prequel. I feel confident saying that since we saw a facehugger implant an alien into a body of an Engineer, then watched it burst out of the Engineer's chest, JUST LIKE IN ALIEN.
If it isn't a prequel, it would make as much sense as Spock whipping out a lightsaber to fight Gary Mitchell/Khan/Whoever in the new Star Trek movie coming out.
