Both are very capable rounds, it really comes down to a question of personal preference. I suggest you develop a ball park idea of what kind of pistol you want (Glock, 1911, etc.) Then find a good range that has rentals, buy a box of .40 and .45 and run some rounds through both the .40 and .45 versions of whatever model, or as close as they have available, you are looking at. I've always felt the recoil on a .40 is a little harsher than a .45, but I've not shot a lot of .45s.

For something local, Cabela's has the best selection by far, and they've always got some good ammo for sale.
