To bring a guy up from the practice squad, you have to cut someone from the 53 man roster. Deactivating someone doesn't clear any room at all.

If you do bring a guy up from the practice squad (after first making room for him by cutting someone), you can certainly fill his spot on the practice squad with someone else.
